Abstract目的:(1)研究94例结直肠癌组织标本中CD133、血管内皮生长因子的表达及其与临床病理特征之间的关系,探讨CD133表达与肿瘤微血管密度(MVD)、VEGF表达之间的相关性。 (2)从结肠癌细胞系SW480中分离CD133+细胞并检测内皮细胞对CD133+细胞表达VEGF的影响。 方法:(1)采用免疫组织化学SP两步法和组织芯片技术检测94例结直肠癌标本中CD133、VEGF、CD31的表达情况。 (2)分别利用免疫磁珠分选技术和无血清悬浮培养法富集CD133+细胞。将SW480细胞或磁珠分选CD133+细胞分别在6种培养条件下进行培养,利用免疫组织化学法检测六组细胞在培养不同时间后CD133及VEGF的表达。 结果:(1)结直肠癌中CD133的表达与肿瘤的分化程度及淋巴结转移有关(P<0.05)。 (2)结直肠癌组织中CD133的表达与VEGF表达及MVD呈负相关关系。 (3)结直肠癌组织中微血管密度按CD133-/VEGF-,+ →CD133+/VEGF-,+ →CD133+/VEGF++,+++ →CD133-/VEGF++,+++的顺序逐渐增加。 (4)在含血清培养基或无血清培养基中,与内皮细胞共培养的SW480细胞CD133+细胞比率与不添加内皮细胞者比有所增加(P<0.05)。 (5)在无血清培养基中,SW480细胞和CD133+细胞添加内皮细胞诱导后CD133+细胞表达VEGF的比率显著增加(P<0.01)。 (6)在培养不同时间后,六组不同培养条件下的细胞中CD133阳性细胞比率和VEGF阳性细胞比率之间呈负相关关系。 结论: (1)结肠癌中内皮细胞可能促进结肠癌干细胞的自我更新。 (2)内皮细胞可能通过诱导结肠癌干细胞表达VEGF来促进血管的发生。 (3)CD133+细胞可能是通过自分泌或旁分泌VEGF的方式促进血管新生,微血管密度增加则促进CD133+细胞的分化成熟。
Other AbstractObjective:(1)To explore the expression of vascular endothelial growth factor and CD133 and investigated their relationship with clinical significances of 94 colorectal carcinoma patients. The expression of CD133 was correlated with MVD and VEGF. (2) Colon cancer stem cell was isolated from the colon cancer cell line SW480 and the effect of endothelial cells on VEGF expression of colon cancer stem cells was investigated. Methods:(1)The expression of CD133,VEGF and MVD(labeled by CD31) were detected by immunohistochemistry using tissue microarrays consisting of 94 specimens of colorectal carcinoma. (2)Serum-free medium under floating culture system and magnetic activated cell sorting techniques were used to enrich or separate CD133+ cells from SW480 cells.The SW480 cells and CD133+ cells had been cultured in six different conditions.The expression of CD133 and VEGF on colon cancer cells were detected by immunohistochemistry. Results:(1)Expression of CD133 was significantly associated with tumor differentiation and lymph node metastasis in colorectal carcinoma(P<0.05). (2)There were significant negative correlations between expression of CD133 ,MVD and VEGF in colorectal carcinoma. (3)In colorectal carcinoma,MVD was gradually increased according to the following order:CD133-/VEGF-,+→CD133+/VEGF-,+→CD133+/VEGF++,+++ →CD133-/VEGF++, +++. (4)When SW480 cells were co-cultured with endothelial cells in serum-supplied medium or serum-free medium,the proportion of CD133+ cells increased significantly(P<0.05). (5)In serum-supplied medium, CD133+cells or SW480 cells co-culture with endothelial cells showed increased expression of VEGF significantly(P<0.01). (6)The proportion of CD133 positive cells was negatively correlated with VEGF in 6 group cells which were cultured for different time. Conclusion:(1)Endothelial cells could maintain the self-renewal and undifferentiated phenotype of CD133+cells. (2)Endothelial cells may promote angiogenesis in the way of inducing SW480 cells to express VEGF. (3) CD133+ cells may promote angiogenesis in colorectal cancer though autocrine- paracrine VEGF and the increasing microvessel density stimulates the differentiation of CD133+ cells.
