党参的糙皮侧耳发酵及发酵液的应用

Abstract本文利用真菌糙皮侧耳生长繁殖能力强、对营养物质要求简单、易培养以及在生长繁殖过程中产生的丰富的酶系有利于对中药材的分解和促进有效成分的释放等特点,对党参进行发酵。首先对糙皮侧耳进行孢子分离,制作糙皮侧耳菌种;然后对党参的发酵条件进行实验研究,采用添加不同浓度党参的PDA平板筛选最佳党参发酵浓度;并分析党参发酵液的成分,同时根据中医药理论对党参发酵液在调节细胞免疫、抗疲劳、抗缺氧、抗血栓、镇痛、拮抗戊巴比妥钠促睡眠作用方面的药理作用进行了初步研究。检测结果表明党参发酵浓度为5%、10%时,糙皮侧耳长势较好。党参发酵后党参皂苷、黄酮含量增加,党参多糖含量减少。HPLC分析检测结果表明发酵前后成分种类及含量有所变化。药理实验结果表明:党参发酵液在调节细胞免疫方面作用不明显;与水煎组相比,适当剂量的党参发酵液在延长小鼠负重游泳时间,提高肝糖原水平方面差异显著,但在降低血清尿素氮含量、血乳酸及提高血乳酸脱氢酶含量方面没有明显差异;与水煎组相比,适当剂量的党参发酵液在延长小鼠断头后张口喘息的存活时间方面差异显著,但在延长小鼠在常压密闭缺氧条件下的存活时间方面没有明显差异;党参发酵液在止血凝血方面与水煎组相比没有明显差异;与水煎组相比,适当剂量的党参发酵液在减少醋酸致小鼠扭体次数方面的作用方面差异显著;适当剂量的党参发酵液在延长小鼠睡眠潜伏期、缩短睡眠时间方面的作用较水煎组有所增强,具有差异显著性。
Other AbstractFungi Pleurotus ostreatus (P. ostreatus) have many fine characteristics, like strongly reproductive Capacity, simple nutriments requirements, easy culture et al. Moreover, the P. ostreatus can produce abundant enzymes that can decompose traditional chinese medicine (TCM) and therefore promote releasing of the effective components from TCM. Thus, in the present work, we used P. ostreatus to ferment Codonopsis pilosula (C. pilosula). Firstly, spore was isolated from P. ostreatus for the preparation of seed. The fermentation condition was optimized with different concentrations of C. pilosula. Secondly, the compositions of fermentation broth were detected and analyzed. Thirdly, the effects of the fermentation broth on regulating the cellular immunity, anti-fatigue, anti-hypoxia, anti-thrombosis, analgesia and antagonizeing the sleep effect of Pentobarbital Sodium were investigated based on the traditional theories. The results showed that P. ostreatus growed well with 5% and 10% fermentation concentration of C. pilosula, it was found that the main medicinal compositions of C. pilosula changed after fermentation. The contents of saponin and flavonoid increased, but the contents of polysaccharide decreased. Both the species and contents of composition changed after fermentation by analyzing with HPLC. Compared with water decoction, animal experiments showed that the proper dose of fermentation broth could significantly prolong swimming time with load of mice and increase liver glycogen, prolong respiration time after decapitation, reduce the mice's twisting caused by acetic acid and enhance the pain threshold induced by heat irritation, prolong sleep latency and shorten sleeping time. However, there were no significant differences in the the mice’s serum urea nitrogen content, blood lactate content, cellular immunity, the levels of lactic dehydrogenase hydrogen, the tolerance to lacking in oxygen under normal pressure, haemostasis, and coagulation between the fermentation broth and the water decoction treatments.
