白阿魏菇深层发酵及其菌丝体多糖生化药理学研究

Abstract本文报告了白阿魏菇Pleurotus eryngii var nebrodensis (=Pleurotus nebrodensis)的深层发酵工艺研究及其菌丝体多糖的提取纯化,结构分析及药理学研究。结果表明:摇瓶发酵的最佳温度为25℃,装液量为三角瓶体积的1/5,培养基的起始pH为5.5,接种量10%,玻璃珠数12个/瓶;最佳碳源为土豆汁;最佳培养基配方为C6H12O60.5%,蔗糖0.5%,玉米粉5%,麸皮4%,蛋白胨0.2%,KH2PO4 0.1%,MgSO4•7H2O 0.05%,羧甲基纤维素钠0.5%,VB1微量;发酵罐发酵时间48h。 菌丝体多糖的提取条件:搅拌下热水浸提3h,pH7.0,温度90℃,料水比1/15,浸提2次;醇析浓度70%;蛋白质去除:样品/氯仿-正丁醇(V/V)为2/1,氯仿/正丁醇(V/V)为2/1,萃取时间40min;得粗多糖(CPNMP),然后经DEAE-cellulose及Sephadex G-100层析柱,得纯多糖(PNMP),纯化后菌丝体多糖纯度为83.22%,气相色谱分析表明PNMP由半乳糖﹑甘露糖﹑葡萄糖和阿拉伯糖等组成;含硫酸基7.5%;糖苷键分别为1→3(主链)和1→6(侧链)。 白阿魏菇菌丝体多糖生物活性:纯多糖(PNMP)能显著清除•OH,抑制•OH所致小鼠肝组织丙二醛的生成和肝线粒体肿胀,减少•OH所致的红细胞溶血,抑制连苯三酚自氧化;粗多糖(CPNMP)可增加巨噬细胞的吞噬活性,增加DTH反应,提高血清中SOD﹑CAT﹑红细胞SOD和溶菌酶活力,因而粗多糖(CPNMP)可以提高机体的免疫力,特别是细胞免疫。粗菌丝体多糖也能显著的抑制二甲苯所致小鼠耳肿胀和毛细血管通透性,增加机体的抗炎性;CPNMP还可以通过免疫系统的作用抑制实体瘤S180的生长,其抑瘤率可达 39.1%。
Other AbstractA study was carried out on the submerged fermented condition and the isolation condition of the mycelium polysaccharide of Pleurotus eryngii var nebrodensis (PNMP). The structure and biological characters of the PNMP and CPNMP were also studied in this paper. The optimum growth temperature, fermentation period, fermentation broth contents of the shake flask culture and initiative pH of Pleurotus eryngii var nebrodensis (=Pleurotus nebrodensis) are 25℃,50mL (250mL of flask) of volume, 10% of volume of inoculation, 12 pearl of glass bead and pH5.5 respectively. The optimum carbon source is potato juice and the optimum fermentation medium is 0.5% glucose, 0.5% sucrose, 5% maize powder, 4% juice bran, 0.2% peptone, 0.1% KH2PO4, 0.05%MgSO4•7H2O,0.05% sodium carboxymethyl cellulose and a little thiamine. The fermentation period of fermenter is 48h. The optimum condition of isolation of mycelium polysaccharide of Pleurotus nebrodensis are 3h and 2 times of isolation, pH7.0, temperature 90℃, rotation, sample/water=1/15, precipitated by ethanol at 70%. Sample/Trichloromthane-Butyl alcohol is 2/1 and Trichloromthane/Butyl alcohol is 2/1 and time is 40 min in removing protein. The contents of total sugar and SO42- were 83.22% and 7.5% separately. PNMP is composed of glucose, galactose, mannose and arabinose. And it has glucosidic bands through 1→3 and 1→6 linkages. PNMP could make ·OH scavenging activity, and inhibit the generation of MDA and self-oxidation. To decrease the swelling of mitochondria and the henmolysis of red blood cells induced by ·OH.. CPNMP could induce activation of PMф and RBC. And enhance their immune functions. CPNMP could markedly decrease tumor weight and promote the phagocytic rate of macrophages and increase weight of immune organ. So the antitumor effects of CPNMP may be related to its immune enhancing activity.
