Abstract | DNA甲基化是一种重要的表观遗传调节方式,在生物的生长发育以及基因的表达调控中起着不可替代的作用。在植物应对自然胁迫的过程中,也有DNA甲基化/去甲基化事件的发生。本研究以拟南芥为材料,研究了DNA去甲基化对植物低温耐受性的影响, 并建立了构建低温胁迫下拟南芥差异甲基化文库的方法。主要结果如下:1.DNA去甲基化能够增加植物的低温耐受性;2.DNA去甲基化增加植物对低温的耐受可能是通过诱导NO产生实现的;3.结合甲基化敏感性限制性内切酶(MS-RE)技术和抑制消减杂交(SSH)技术,构建了低温胁迫下拟南芥差异甲基化文库,来寻找通过甲基化机制进行表达调控的基因,并对其进行分析验证。 |
Other Abstract | DNA methylation/ demethylation occurs when plants are exposed to environment stresses. In the present study, we used Arabidopsis to study the role of DNA demethylation in response to low temperature stress. In order to identify genes regulated by methylation mechanisms under cold stress, we developed a protocol which combines MS-RE digestion with SSH to construct a differential-methylation subtractive library. The main results were summarized as follows: 1.DNA demethylation treatment resulted in increased tolerance to low temperature in Arabidopsis seedling.2.NO production may involve in the increase of tolerance to low temperature resulted from DNA demethylation treatmen.3.We constructed a differential-methylation subtractive library of Arabidopsis in response to low temperature stress by combining MS-RE digestion with SSH for identifying genes regulated by methylation mechanism under cold stress. |
