Abstract目的:检测人急性白血病(AL)骨髓单个核细胞中自噬相关基因Beclin1和微管相关蛋白质轻链3(MAPLC3)的表达,并探讨其意义。方法:采用透射电镜(TEM)和 RT-PCR 方法检测27例初治组、8例难治复发、19例完全缓解(CR)的急性白血病患者和31例正常人骨髓单个核细胞(BMMNC)中的自噬活性以及Beclin1、MAPLC3 mRNA 的表达。结果:透射电镜观察初治组骨髓单个核细胞自噬活性80%;难治复发组骨髓单个核细胞自噬活性100%;完全缓解组骨髓单个核细胞自噬活性40%;正常对照组骨髓单个核细胞自噬活性20%。初治组、难治复发组与正常对照组自噬活性比较差异均有统计学意义(χ2=8.22, P=0.02; χ2=8.17, P=0.00);完全缓解组与正常组自噬活性比较差异无统计学意义(χ2=5.16, P=0.08)。RT-PCR 检测Beclin1 mRNA 在初治组骨髓单个核细胞中相对表达量为0.68±0.18;在难治复发组骨髓单个核细胞中相对表达量为0.79±0.09,完全缓解骨髓单个核细胞细胞中相对表达量为0.52±0.15;正常对照组骨髓单个核细胞中相对表达量为0.57±0.13。初治组、难治复发组与正常对照组比较差异有统计学意义(F=7.05,P=0.01;F=53.63,P=0.00);完全缓解组与正常对照组比较差异无统计学意义(F=1.57,P=0.22)。RT-PCR 检测MAPLC3 mRNA在初治组骨髓单个核细胞中相对表达量为0.24±0.06;在难治复发组骨髓单个核细胞中相对表达量为0.30±0.07;完全缓解骨髓单个核细胞细胞中相对表达量为0.16±0.04;正常对照组骨髓单个核细胞中相对表达量为0.16±0.05。初治组、难治复发组与正常对照组比较差异有统计学意义(F=34.26,P=0.00;F=71.30,P=0.00);完全缓解组与正常对照组比较差异无统计学意义(F=0.40,P=0.53)。
Other AbstractObjective The expression of autophagy-related genes Beclin1 and microtubule-associated protein 1 light chain 3 (MAPLC3) in bone marrow mononuclear cells (BMMNC) isolated from acute leukemia (AL) patients was detected and its significance discussed. Methods Transmission electron microscopy (TEM) and RT-PCR were used to detect autophagy activity and Beclin1, MAPLC3 mRNA expression level in BMMNC isolated from 27 AL patients in de novo, refractory or relapse and completely remission (CR) stages and 31 normal persons. Results The results showed that autophagic activity and the Beclin1, MAPLC3 mRNA expression level in BMMNC from de novo AL patients were 80%, 0.68±0.18, 0.24±0.06 respectively; those in BMMNC from refractory or relapse patients were 100%, 0.79±0.09, 0.30±0.07 respectively; those in BMMNC from CR patients were 40%, 0.52±0.15, 0.16±0.04 respectively; while those in BMMNC from normal control were 20%, 0.57±0.13, 0.16±0.05 respectively.The autophagic activity and the Beclin1, MAPLC3 mRNA expression level in de novo and refractory or relapse patients were higher than those in normal control with statistical significance (p<0.05), while the comparison between CR patients and normal control showed no statistical difference(p>0.05).
