Abstract | 目的:观察脂联素对人脐静脉内皮细胞(HUVECs)补体调节蛋白CD55和CD59表达的影响。
方法:体外培养人脐静脉内皮细胞;分为空白对照组、C-反应蛋白(CRP)组和脂联素组,人脐静脉内皮细胞分别与50ug/ml的CRP和25ug/ml的脂联素共同孵育48小时,流式细胞仪检测CD55和CD59的表达。
结果:CRP刺激后CD55平均荧光强度、CD59平均荧光强度和CD55阳性百分率较空白对照组升高(3.84±0.21比2.81±0.06,p<0.001;69.72±4.48比51.15±7.86,p<0.01;98.30±1.13比93.57±1.18,p<0.001);脂联素刺激后CD59平均荧光强度和CD55阳性百分率较空白对照组升高(63.02±4.90比51.15±7.86,p<0.05;96.55±0.66比93.57±1.18,p<0.01)。
结论:脂联素是人脐静脉内皮细胞CD55和CD59表达的上调因素,并可能通过上调内皮细胞CD55和CD59的表达发挥抗炎、抗动脉粥样硬化作用。 |
Other Abstract | Object To observe influence of adiponectin on expression of CD55 and CD59 which expressed on human umbilical vein endothelial cells(HUVECs).
Methods Cultivate HUVECs in vitro. HUVECs were divided into three groups--–CRP, adiponectin and contral.HUVECs were respectively incubated with 50ug/ml CRP and 25ug/ml adiponectin for 48 hours.CD55 and CD59 expression on HUVECs were detected by flow cytometry.
Results CD55 mean fluorescence intensity,CD59 mean fluorescence intensity and CD55 percentage were increased versus contral after CRP stimulated (3.84±0.21 versus 2.81±0.06,p<0.001;69.72±4.48 versus 51.15±7.86,p<0.01;98.30±1.13 versus 93.57±1.18,p<0.001) .CD59 mean fluorescence idensity and CD55 percentage were increased versus contral after adiponectin stimulated(63.02±4.90 versus 51.15±7.86,P<0.05;96.55±0.66 versus 93.57±1.18,P<0.01).
Conclusion Adiponectin can upregulates expression of complement regulatory protein CD55 and CD59,and may contributes to anti-inflammation and anti-atherosclerosis by this response. |
