Abstract目的 观察水通道蛋白1,4(AQP1,4)在正常大鼠角膜和晶状体生长发育过程中的表达及分布。确定2种水通道蛋白在角膜和晶状体的初次表达时间、部位和表达趋势。 方法 用免疫组织化学方法观察水通道蛋白1,4分别在胚胎13天、15天、18天、20天、出生当天、出生后7天、14天和8周成年大鼠角膜及晶体中的表达。每组取6只健康大鼠,(共48只大鼠),分别进行HE染色和免疫组织化学染色。 结果 AQP1在胚胎15天大鼠的角膜内皮和晶体上皮就有表达,胚胎18天表达增强,从胚胎20天开始趋于稳定。角膜上皮始终未见AQP1表达。AQP4在生后7天大鼠的角膜上皮、晶体上皮和晶体纤维有弱阳性表达,生后14天表达增强并持续到成年鼠。角膜内皮始终未见AQP4表达。 结论 AQP1,4在大鼠角膜和晶体的表达时间、部位和趋势不同。AQP1,4的表达随着大鼠角膜和晶体的生长发育呈动态变化。推测此2种蛋白对维持角膜和晶体的透明性及正常生理功能发挥了重要作用。 关键词 水通道蛋白-1;水通道蛋白-4;角膜;晶状体;免疫组织化学
Other AbstractObjective To observe the expression and distribution of AQP1 and AQP4 during development of normal rat cornea and lens.In order to confirm the emergence, location and tendency of AQP1 and AQP4 in rat cornea and lens. Methods cornea and lens of eyes dissected from embryonic day (E)13,E15,E18,and E20,postnatal day (P)0,P7,and P14,and postnatal week 8 rats were subjected to Immunohistochemical technique with AQP isoform-specific antibodies. Resuits Developmentally,AQP1 was detected as early as E15 in immature corneal endothelium and lens epithelial cells.The period of E18 expressed enhancer.The period from E20 to P14 and adult expressed steadily in the rat development.However, corneal epithelial cells did not expressed. In contrast,AQP4 was first observed at P7 in the developing cornea and len.It expressed strong in P14 and expression persisted throughout development up to adulthood. However, AQP4 did not expressed in corneal endothelium. Conclusion The emergence, location and tendency of AQP1 are different from AQP4.expression of AQP1 is dynamic along with cornea and lens development.These two proteins are important to keep physiology balance and transparency of cornea and lens.
