| Abstract | 目的：
观察p-AKT（磷酸化蛋白激酶B）和p-Mtor(磷酸化哺乳动物雷帕霉素靶蛋白)因子在肾透明细胞癌组织中的表达，为mTOR及AKT抑制剂应用于肾透明细胞癌的治疗提供依据。
方法：用免疫组化染色测定肾透明细胞癌和正常肾组织中p-AKT和p-mTOR 的表达情况。
结果:p-AKT在肾透明细胞癌和正常肾组织中的阳性表达率分别为61.3%、26.1%(P<0.01），p-mTOR的阳性表达率分别为56％、28.3％（P<0.05）。p-AKT表达与肿瘤临床分期明显相关，临床分期越高，其阳性表达率越高（P=0.029<0.05），p-mTOR蛋白表达与肿瘤临床分期也具有相关性（P=0.043<0.05）。p-AKT和p-mTOR表达与肾透明细胞癌病理分级密切相关（p-AKT：P=0.040<0.05；p-mTOR：P=0.014<0.05）。肾透明细胞癌组织中p-AKT与p-mTOR 的表达呈正相关（r=0.953，P=0.000）。
结论：AKT/mTOR 信号通路在肾透明细胞癌组织中过度激活，在肾透明细胞癌发生发展中起到重要作用。理论上，应用mTOR及AKT抑制剂可治疗肾透明细胞癌。 |
| Other Abstract | Objective This study was to explore the expression of p-AKT and p-mTOR factors in clear cell carcinoma of kidney and to provide evidence for mTOR and AKT inhibitors used in renal clear cell carcinoma therapy.
Methods The expression of p-AKT and p-mTOR in clear cell carcinoma of kidney and normal kidney tissue were measured by immunohistochemical method.Results The level of p-AKT p-mTOR and expression in renal clear cell carcinoma were significantly higher than in normal kindney tissues(p<0.01).The expression of p-AKT significantly associated with tumor clinical stage and the higher the clinical stage,the higher the rate of positive expression of p-AKT (P = 0.029 <0.05).The expression of p-mTOR protein was relative with tumor clinical stage (P = 0.043 <0.05).The expressions of p-AKT and p-mTOR had close correlation with pathological grading（p-AKT：P=0.040<0.05；p-mTORP=0.014<0.05）.The expression of p-AKT in renal clear cell carcinoma was positively correlated with p-mTOR(r = 0.953,P = 0.000).
Conclusion AKT/ mTOR signaling pathway was over- activated in renal clear cell carcinoma.They may play an important role in occurrence and development of renal cell carcinoma.In theory,the mTOR and AKT inhibitors can be used to treat the renal cell carcinoma. |
