Abstract目的 探讨深度血液稀释(profound homedilution)对大鼠T reg细胞及组织代谢的影响。方法 健康雄性SD大鼠20只,体重350~400g,戊巴比妥腹腔注射麻醉后,解剖颈总动脉连接L-420E动物测压系统监测血压(BP)和心率(HR),并用于放血和测定血气。解剖股静脉后留置24G套管针,备采血和输液[6%贺斯(HES,200/0.5)和携氧液]用,输液用微量注射泵泵入,先输贺斯后输携氧液,两者之比为1:2。当Hct值达目的值(<20%)时,静脉输液停止。分时段T(稀释前)、T(稀释后即刻)取静脉血各1ml,置ADTA-K抗凝管中送流式细胞仪检测CD、CD;稀释结束后24小时,处死动物,取动物心、肺、肝、肾,行光镜下组织病理学观察。结果 其中三只大鼠被剔除(实验初期一只窒息死亡,另两只标本凝固),其余17只Hct值达到目标值(17.41±1.28) %且动物全部存活。输液量(30.00±5.00)ml,动物放血量(10.00±2.00)ml,用时(60.00±10.00)min;血液稀释后CD+CD+、氧分压显著升高(P<0.01),病理切片显示组织无缺血缺氧改变及水肿表现。结论 应用携氧液行深度血液稀释,可使大鼠外周血中Treg细胞及血氧分压明显升高,且血流动力学稳定,组织无缺血缺氧及水肿改变。
Other AbstractObjective To study the effects of profound hemodilution with oxygen-carrying liquid on regulatory T cells and tissue metabolism of rats. Methods Twenty male adult SD rats weighting 350~400g were used in this study. The animals were anesthetized with intraperitoneal pentobarbital. Arteria carotis communis and femoral vein were cannulated for HR and BP monitoring. Blood was withdrawn from artery and simultaneously replaced by intravenous infusion of equal volume of 6% HES and oxygen-carrying liquid until hematocrit was 10% ~20%. Blood samples were taken before hemodilution and end of hemodilution for determination of regulatory T cells by flow cytometer and analysis blood gas. After twenty-four hours, the animals were killed and the heart,lung,liver and kidney of animals were gained to put into the container with formalin, the materials were performed pathological section. Results There were seventeen rats were survival and hematocrit reached objective value(17.41±1.28) %. Hematocrit was significantly attenuated(P<0.01), CD4+CD25+ and oxygen partial pressure were significantly increased(P<0.01), the pathological section disclosed that the tissues were non-anoxyaemia and non-edema.Conclusion The regulatory T cells of rats could be significantly increased through profound homedilution, haemodynamics were tabilization and the tissues were non-anoxyaemia and non-edema, The oxygen partial pressure of rats may significantly higher by using oxygen-carrying liquid .
