Abstract目的: 三磷酸腺苷(adenosine triphosphate,ATP)能促进脊髓损伤(spinal cord injury,SCI)的修复,通过观察ATP联合骨髓间充质干细胞(bone marrow mesenchymal stem cells, BMSCs)移植治疗大鼠SCI的疗效,探讨两者在SCI修复中的协同作用及其联合移植治疗SCI的可行性。 方法:取1只体重为120 g的雄性SD大鼠的股骨和胫骨骨髓,分离、培养获得BMSCs。将第3代细胞BrdU标记,制备浓度为5.0×107个/ mL的细胞悬液。将48只体重240~260 g 的SD雌性大鼠,采用改良Allen’s打击法制备T11~13节段SCI模型后,随机分为4组,每组12只。联合移植组(A组):将ATP(40 mg/kg)与6μl BMSCs细胞悬液混匀后,注射至损伤脊髓撞击中央及距头、尾侧各1 mm处,细胞数量约3.0×105个;BMSCs移植组(B组)、ATP移植组(C组)及对照组(D组):同A组方法分别移植BMSCs细胞悬液6 μl、ATP(40 mg/kg)及PBS(40 mg/kg)。术后观察大鼠一般情况,于1、3、7、14、28 d采用Rivilin斜板实验及Tarlov评分评价后肢功能恢复程度;28 d处死后取材行组织学和免疫组织化学染色观察脊髓损伤修复情况。
Other AbstractObjective: Adenosine triphosphate(ATP) can promote the repairation of spinal cord injury (SCI). To investigate the effect of acute spinal cord injury repaired by ATP combing with bone marrow mesenchymal stem cells (BMSCs) transplantation, and to evaluate the synergistic action of ATP and BMSCs in the repairation of SCI and the feasibility of the combined transplantation in the treatment of SCI. Methods BMSCs were isolated from a male SD rat (weighing 120g) using the marrow of the tibia and the femur, the third generation BMSCs were proliferated and labeled with BrdU, then made into BMSCs suspension of 5.0×107/ mL. 48 adult female SD rats(weighing 240~260g) were used to make acute SCI models at T11~13 level according to the improved Allen’s method, were randomly divided into 4 groups(groups A, B, C and D, n=12). the group of ATP-BMSCs transplantation(group A): Inject ATP(40mg/kg) and BMSCs(6μl) to the sentral point and the other two points which were 1mm from the each side of head and tail of the injured spinal cord after blending the BMSCs suspension, the cells amount was about 3.0×105; BMSCs transplantation(group B), ATP treatment(group C) and PBS control(group D): Transplant the BMSCs suspension(6μl) , ATP(40mg/kg) and PBS(40 mg/kg) to the points as the metods of group A. Observated the general conditions of the rats after opration; The nerve function of low extremities was evaluated using the promoted Tarlov scale and the Rivilin inclined plane test at 1, 3, 7, 14, 21 and 28 days after transplantation; All the rats were execute 28 days after operation, the reparative effect of SCI was observated using histological and immunohistochemical staining.
