Abstract | 目的 分析0~3岁三聚氰胺致婴幼儿泌尿系结石的发病情况及特点,研究三聚氰胺在人体及动物体内的代谢过程。
方法 将120例诊断为三聚氰胺肾结石的婴幼儿按照停用奶粉时间分为三组:停用奶粉时间>5天,停用奶粉时间<5天,停用奶粉时间<2天,每组40例。用高效液相色谱(HPLC)检测120例婴幼儿血、尿中的三聚氰胺。将20只大鼠随机分为两组,每组10只,两组每天分别以300mg/Kg和100mg/Kg剂量的三聚氰胺灌胃,持续10天灌胃后于2小时、5小时、10小时、24小时、48小时收集大鼠血标本,用HPLC检测20只大鼠血标本中三聚氰胺的含量。
结果 120例三聚氰胺肾结石患儿停用三聚氰胺污染奶粉以后在血液中均未检测到三聚氰胺,在40例停用奶粉时间>5天的患儿尿液中未检测到三聚氰胺,在40例停用奶粉时间<5天的患儿尿液中有12例检测到三聚氰胺,在40例停用奶粉时间<2天的患儿尿液中有25例检测到三聚氰胺。大鼠灌胃2小时血液中三聚氰胺浓度最高,5小时血液中三聚氰胺浓度小于2小时浓度的一半,10小时后血液中三聚氰胺含量很低,24小时后血液中已检测不到三聚氰胺。
结论 三聚氰胺在婴幼儿体内及大鼠体内的代谢速度很快,24小时后在大鼠体内已基本清除,48小时后在人体血液中已检测不到。三聚氰胺在人体及大鼠体内以原形经尿排出体外。 |
Other Abstract | Objective To indentify the characteristic of urolithiasis in infants aged 0-3 years old fed by Melamine-tainted Milk Powder in Gan Su province and study metabolism of melamine in human and rat .
Method 120 cases diagnosed as kidney stone were divided into 3 groups due to discontinuity of Melamine-tainted Milk Powder and each group contained 40 cases: the discontinuity of group 1 exceeded more than 5 days, group 2 was between 2 and 5 days and the discontinuity of group 3 was less than 2 days. Melamine in serum and urine was detected by High Performance Liquid Chromatography (HPLC) for these cases. 20 rats were divided into two groups and every group contained 10 cases, group 1 was intervened with intragastric administration by the dose of 300mg/Kg and group 2 was intervened with intragastric administration by the dose of 100mg/Kg.10 days later, the blood of SD rats were collected in 2h, 5h,10h,24h,48h after intragastric administration and Melamine in serum was also detected by High Performance Liquid Chromatography (HPLC) .
Results In blood samples, melamine was undetected after discontinuity of Melamine-tainted Milk Powder in all of 120 cases diagnosed as kidney stone. In urinary samples melamine was undetected in all of 40 cases in group 1.In group 2, melamine was detected in 12 urinary samples of all 40 cases. In group 3, melamine was detected in 25 urinary samples of all 40 cases. In rats body, the levels of melamine in blood was higher in 2 hours, the levels of melamine in 5 hours was less than the half of that in 2 hours, the levers of melamine in 10 hours was lower and after 24 hours melamine was almost undetected in blood.
Conclusion The metabolism of melamine in human and rat are very quickly, it will be cleared up in 24 hours in rat , and will be not detected in blood for human after 48 hours, and then will be discharged from the urine in the original shape. |
