Abstract利用CCK-8法检测黄芪多糖对HL-60、K562细胞的半数抑制量(IC50),以此含量作用HL-60、K562细胞48小时后,采用RT-PCR、流式细胞仪分别检测作用前后白血病细胞MICA基因和蛋白水平的表达变化。运用乳酸脱氢酶释放法检测NK细胞对黄芪多糖处理前后HL-60、K562细胞的杀伤率。 结果:经黄芪多糖处理后HL-60细胞MICA mRNA表达是处理前的1.46±0.16倍,在不同效靶比(5:1、10:1、20:1)下均有显著差异,差异有统计学意义( P<0.05) 。 结论:1.黄芪多糖作用后白血病细胞HL-60、K562的MICA mRNA及MICA表达均显著上调;2.黄芪多糖能可以通过上调白血病细胞MICA的表达增强白血病细胞对NK细胞的杀伤敏感性。
Other AbstractObjective: This study based on the observation of that if astragalus polysaccharides could increase the expression of MICA in HL-60, K562 cells and enhance the cytotoxicity of NK cell against leukemia cell lines, and to find new targets for NK cell-based immunotherapy in leukemia. Methods: The different concentrations of astragalus polysaccharides incubated with the HL-60, effector-to-target cell ratios (E:T) before and after treating with APS. Results: The MICA mRNA of HL-60 cells after treating with APS is 1.46±0.16 times than that of before treating with APS; MICA mRNA of K562 cells after treating with APS is 2.26±0.34 times than that of before ( P < 0.05). the difference have statistical significance ( P< 0.05). Conclusion: The APS could enhance the sensitivity of leukemia cells to NK cell through the up-expression of MICA on the surface of leukemia cell.
