Abstract目的 探讨黄连素(berberine)对人卵巢癌SKOV3细胞增殖的抑制作用及其诱导细胞凋亡和对错配修复基因hMLH1启动子区CpG岛的去甲基化作用。 方法 MTT法检测berberine对人卵巢癌SKOV3细胞增殖的抑制作用;流式细胞术(FCM)检测berberine作用前后细胞周期分布的改变和细胞凋亡率的改变。 结果 berberine对人卵巢癌SKOV3细胞增殖有较强的抑制作用,呈浓度和时间依赖性,berberine与顺铂联合使用,顺铂IC50值明显下降,组间比较差异有统计学意义(P<0.05);berberine作用后,G0/G1期细胞增多(P<0.05),细胞凋亡率明显增加(P<0.05),并出现典型的凋亡细胞形态学和超微结构的改变;hMLH1基因启动子区CpG岛在SKOV3细胞部分甲基化,berberine作用后,hMLH1基因启动子区CpG岛甲基化特异性条带减弱,非甲基化特异性条带增强;berberine作用后,bcl-2、survivin基因mRNA表达下调,bax、hMLH1基因mRNA表达上调。 结论 berberine抑制人卵巢癌SKOV3细胞的增殖作用可能是通过诱导细胞凋亡和逆转hMLH1 基因启动子区CpG岛的甲基化来实现的,berberine和顺铂联合使用,可增加化疗敏感性。
Other AbstractObjective To investigate the effect of berberine on inhibition of proliferation through induction of apoptosis and the demethylation on mismatch repair gene hMLH1 in human ovarian cancer SKOV3 cells. Methods The inhibitory effect of berberine on the proliferation of human ovarian cancer SKOV3 cells was evaluated by the MTT assay. Results Berberine significantly inhibited the proliferation of human ovarian cancer SKOV3 cells in a dose-dependent and time-dependentmanner, IC50 of cisplatin dropped obviously when combined with berberine, the differences were significant among different groups (P<0.05)
