Abstract骨形态发生蛋白在鼠角膜成长过程中的表达 目的:确定转化生长因子-β(transforming growth factor TGF-β)超家族成员转化生长因子-β2及骨形态发生蛋白-2(bone morphogenetic proteins BMP-2)在正常大鼠角膜生长发育过程中的表达及作用。 方法:本实验将Wistar大鼠分为六个年龄组:即为胚胎17天(E17),出生当天(P0),出生后4,7,10天和11周(用P4, P7, P10, and 11w表示)六组,每个年龄组包括六只大鼠。分别于以上年龄段摘除大鼠眼球,用免疫组织化学方法观察TGF-β超家族成员转化生长因子-β2和骨形态发生蛋白-2分别在大鼠胚胎17天(E17),出生当天(P0),出生后4,7,10天和11周(P4, P7, P10, and 11w)角膜蛋白水平的表达。 结果:转化生长因子-β2和骨形态发生蛋白-2在大鼠胚胎17天时就有表达,于出生当天(P0)至出生后10天(P10)及成年大鼠(P11W)角膜生长发育过程中呈动态变化过程。TGF-β2于E17、P4、P7、P11W期角膜各层均有表达,P0、P10期各层呈弱阳性表达。BMP-2则于E17、P0、P7、P10、P11W期角膜各层均有表达,P4期角膜各层呈弱阳性表达。 结论:本实验检测到TGF-β超家族成员TGF-β2、BMP-2在大鼠角膜的表达随着角膜的生长发育而成动态调节过程。结果提示TGF-β2、BMP-2与角膜的胚胎期生长发育及维持出生后角膜的动态平衡有关, BMP-2可能也参与组成角膜细胞因子网络调节角膜细胞的增殖和分化。 "
Other AbstractExpression of bone morphogenetic proteins in Developing rat Cornea Purpose: To ascertain the role played and the expression of the transforming growth factor (TGF)-βsuperfamily TGF-β2 and BMP-2( bone morphogenetic proteins -2)during development of the normal rat cornea. Methods: Wistar rats were used in this study. They had been divided into six groups by age.The eyes of the Wistar rats were enucleated at embryonic age 17 days (E17) and on 0, 3, 6, and 9 days, and 11 weeks postnatally (designated as P0, P3, P6, P9, and adult(P11W), respectively). Six rats at each age were sacrificed for observation. Immunohistochemistry were used to examine .The expression of the TGF-βsuperfamily TGF-β2 and BMP-2 were observed at the protein level in rat cornea at embryonic age 17 days (E17), at birth (P0), at postnatal days 4,7, 10, and at 11 weeks(P4,P7,P10,and adult(P11W) respectively). Results:TGF-β2 and BMP-2 were detected at E17. The period from P0 to P10 and adult(P11W)corresponded to the period of dynamic changes in the normal rat cornea development. TGF-β2 were detected at E17、P4、P7、P11W throughout the rat cornea and were decreased at P0、P10. BMP-2 were detected at E17、P0、P7、P10、P11W throughout the rat cornea and were decreased at P4. Conclusion: The results suggest that the expression of TGF-βsuperfamily TGF-β2 and BMP-2 is regulated along with corneal development and may be related to corneal development and maintain the homeostasis after birth. the results suggest that BMP-2 may be components of the corneal cytokine network and may participate in the regulation of cellular proliferation and differentiation
