Abstract目的:通过检测血小板反应蛋白-1(Thrombospodin-1, TSP-1)在骨肉瘤中的表达,探讨其与骨肉瘤组织血管生成、侵袭性的关系。 方法:收集兰州大学第一医院和甘肃省肿瘤医院2003年1月~2007年2月手术切除54例经临床、病理、X线相结合确诊为骨肉瘤的石蜡包埋标本,通过免疫组织化学链霉抗生物素蛋白-过氧化物酶(S-P) 法检测骨肉瘤组织中TSP-1的表达,用CD34单克隆抗体标记血管内皮细胞计数骨肉瘤MVD,经OLYMPUM研究级荧光数码照相(DP71)显微镜(BX51)读取图像并放大输入计算机,所得数据输入SPSS13.0,分析TSP-1各表达组与MVD计数的关系及其临床病理学意义。 结果:54例骨肉瘤组织中,TSP-1的阳性表达率为33.3%(18/54)。骨肉瘤TSP-1的表达程度与MVD呈负相关,TSP-1高表达组的MVD明显低于TSP-1低表达组;骨肉瘤TSP-1表达与骨肉瘤临床病理因素无统计学意义,但与肺转移显著相关。 结论:TSP-1在骨肉瘤发生发展中起重要作用,并影响血管形成及肿瘤侵袭性,有望成为治疗骨肉瘤的新靶点。
Other AbstractOBJECTIVE: To explore the expression of Thrombospodin-1(TSP-1) in osteosarcoma and investigate its relationship between the expression and invasion. microvessel density of adenomas. METHODS: 54 cases with osteosarcoma tissue specimens were obtained from the pathology department of Lanzhou University First Hospital and Gansu Provincial Tumor Hospital between January 2003 and February 2007, fixed in 4% paraformaldehyde, embedded in paraffin. The expression of TSP-1 was evaluated by immunohistochemical staining of tumor speciments from 54 patients with osteosarcoma. The microvessel density(MVD) was assessed by immunostaining with CD34 in the most neovascularized area(hot spots). Images were read and enlarged by research-level digital camera fluorescence microscope and entered into the computer.The data were entered into SPSS13.0 to proceed statistical analysis to find out the relationship between angiogenesis and TSP-1 expression in osteosarcoma and their clinicopathological significance. RESULTS: The positive rate of TSP-1 in 54 osteosarcoma specimens was 33.3%(18/54). In osteosarcoma, the mean microvessel count of TSP-1 trong-expression tumors was significantly lower than that of TSP-1 low-expression tumors.The close correlation was found between TSP-1 expression and microvessel density. TSP-1 expression and microvessel density was remarkably correlated with lung etastasis in osteosarcoma. In contrast, no association was found between TSP-1 epression and gender, age, size and location of tumor, osteosarcoma classification, as well as microvessel density. CONCLUSION: TSP-1 might play an important role in the evelopment of osteosarcoma,and TSP-1 regulates invasiveness and ngiogenesis.Thus, TSP-1 might be a newtherapy target in osteosarcoma.
