Abstract目的:1.了解大鼠骨髓间充质干细胞生物学特性及归巢特征;2.通过重组腺病毒介导的人肝细胞生长因子基因转染骨髓间充质干细胞,探讨其转染效率、目的基因的表达情况;3.探讨携带有肝细胞生长因子基因的骨髓间充质干细胞对移植颗粒脂肪的新生血管和存活率的影响。 结论:1.骨髓间充质干细胞体外能向成脂分化;导入目的基因不会改变MSCs归巢特性;2. 重组腺病毒可介导肝细胞生长因子基因转染骨髓间充质干细胞,有效表达目的蛋白;3.携带HGF基因的MSCs,在移植体中持续表达HGF,有效的促进移植颗粒脂肪的新生血管形成;减少颗粒脂肪移植后坏死、吸收,从而提高移植脂肪的存活率。
Other AbstractObjective: 1.To study the biological and homing characteristics of mesenchymal stem cells. 2. To discuss the transfer efficiency of recombinant adenovirus-mediated green fluorescent protein(Ad-GFP) to MSCs and HGF expression in MSCs carrying human hepatocyte growth factor gene (Ad-HGF). 3.To investigate the effect of MSCs transfected with HGF gene on the angiogenesis and the survival rate of free grain fat grafts. Conclusion: 1. MSCs would be induced into adipose cells ; Transfection of target DNA into MSCs probably might not change their homing characteristics. 2. The recombinant adenovirus carrying human hepatocyte growth factor gene (Ad-HGF) would transfect MSCs. The transfected cells could efficiently express the target gene. 3. MSCs transfected with Ad-HGF would express HGF, promote angiogenesis in grain fat grafts, and increase the survival rate of fat grafts.
