Abstract目的:探讨TGR5和FXR在结直肠癌患者组织的表达及临床意义。 方法:收集2011年6月至2011年12月结直肠癌患者手术标本36例,以癌旁组织作为对照,应用荧光定量PCR(real time quantity PCR RT-PCR)检测结直肠癌组织、癌旁组织和正常组织中TGR5和FXR的表达水平,分析TGR5和FXR表达与结直肠癌临床病理参数及代谢相关疾病和代谢相关生化指标之间的关系。 结果:荧光定量RT-PCR结果显示;结直肠癌组织中TGR5mRNA的表达水平为11.97±6.39, 癌旁组织表达水平为5.61±3.62,明显高于正常组织,(P均<0.05), 癌组织高于癌旁组织(P<0.05),各组间差异有统计学意义(P<0.05)。结直肠癌组织中FXRmRNA的表达水平为0.164±0.347, 癌旁组织表达水平为0.375±0.314,癌组织低于癌旁组织(P<0.05),癌旁组织低于正常组织(P<0.05),各组间差异亦有统计学意义(P<0.05);Ⅰ期、Ⅱ期、Ⅲ期、Ⅳ期结直肠癌组织TGR5和FXRmRNA的表达水平有进行性升高和下降的趋势,但差异无统计学意义(P>0.05);高、中、低分化结直肠癌组织中TGR5和FXRmRNA的表达水平呈进行性升高和降低,但差异无统计学意义(P>0.05)。 结论: 1. TGR5mRNA在结直肠癌和癌旁组织中表达升高。2. FXRmRNA在结直肠癌和癌旁组织中表达下降。
Other AbstractObjective To investigated the expression of TGR5and FXR in human colorectal cancer and its clinical significance. FXR expression was not associated with metastasis and without metastasis respectively. the difference was not statistically significant (P>0.05). Conclusions TGR5 and FXR was higher and lower expression in human colorectal cancer than normal colorectal tissues respectively, might be associated with the genesis and development of colorectal cancer.
