大鼠颈动脉球囊损伤后血清VEGF、TNF-α和IL-6的变化及其干预

Abstract摘要: 目的 探讨大鼠颈动脉球囊损伤后血清血管内皮生长因子(VEGF)、肿瘤坏死因子(TNF-α)和白介素-6(IL-6)水平及其干预。 方法 60只健康雄性SD大鼠随机分为对照组(A组)、模型组(B组)、沙利度胺小剂量组(C组)和沙利度胺大剂量组(D组)。A组大鼠颈动脉没有被球囊损伤;B组大鼠颈动脉被球囊损伤;C组大鼠自球囊损伤后给予100 mg•kg-1•d-1沙利度胺灌胃给药;D组大鼠自球囊损伤后给予200 mg•kg-1•d-1沙利度胺灌胃给药。分别于术后7d、14d 和28天处死大鼠,用酶联免疫吸附法( ELISA)检测血清中VEGF浓度;采用放射免疫法检测血清中TNF -α和IL -6的浓度。 结果 与A组比较,B组、C组和D组血清VEGF、TNF-α和IL-6浓度在损伤后整个实验中均明显高于A组水平(P < 0. 05);A组、C组和D组大鼠血清VEGF、TNF -α和IL-6浓度在损伤后不同时间点明显低于B组(P < 0. 05);与C组比较,D组VEGF、TNF -α和IL-6浓度在损伤后不同时间点均明显降低(P < 0. 05)。 结论 大鼠颈动脉球囊损伤后血清VEGF、TNF-α和IL-6水平明显升高;沙利度胺可有效降低大鼠颈动脉球囊损伤后血清VEGF、TNF-α和IL-6水平,且降低水平与沙利度胺剂量有关。 关键词:大鼠;肿瘤坏死因子;白介素-6;血管内皮生长因子;球囊损伤
Other AbstractObjective To investigate the Levels and Effect of Vascular endothelial growth factor (VEGF)、tumor necrosis factor-α(TNF-α) and InterLeuk- in-6(IL-6) after balloon injury in rat carotid artery. Methods 60male Sprague-Dawley rats were divided into the control group(A,no injury) 、the model group(B,the simple injury)、thalidomide small doses group(C,the simple injury,100 mg•kg-1•d-1 of thalidomide was administered after conventional carotid artery denudation injury)and the high doses group(D,the simple injury,200 mg• kg-1•d-1 of thalidomide was administered after conventional carotid artey after arten injury).The rats were killed at 7、14、28days. Serum concentrations of VEGF were measured by enzyme–linked immunosorbent assay (ELISA) and serum concen- trations of TNF -α、IL-6 were measured radioimmunoassay immediately. Results Serum concentrations of VEGF、TNF -αand IL-6 were significantly higher in B、C and D group throughout the experiment in comparison with A group ( P < 0.05); Compared with the B group,serum concentrations of TNF -α、IL-6 and VEGF were markedly reduced in A、C and D group in different times after injury (P < 0.05);Compared with the C group,Serum concen- trations of VEGF、TNF -αand IL-6 were also markedly reduced in D group in different times after injury (P < 0.05). Conclusions TNF -α、IL-6 and VEGF were high expressed obviously after arterial injury. We found that thalidomide, effectively inhibits Serum IL-6,TNF -αand VEGF in balloon-injured rat carotid arteries.The level of three factors were correlated with the dose of thalidomide.
