|Abstract||目的：评价川芎嗪对慢性坐骨神经压迫性损伤大鼠的脊髓背根神经节GAP- 43 mRNA表达的影响，初步探讨川芎嗪对神经病理性疼痛大鼠的作用和相关的机制。
方法：选取成年雄性健康Wistar大鼠共54只，周龄6~8周，体重180~220 g。分组方法采用随机数字表法，即将54只大鼠分为3组（n=18）：假手术组（S组）、神经病理性疼痛组（NP组）和川芎嗪组（TMP组）。NP组和TMP组采用慢性坐骨神经压迫损伤法建立神经病理性疼痛模型，S组大鼠仅暴露坐骨神经不结扎。TMP组于CCI术毕即刻腹腔注射川芎嗪100 mgkg-1，1天1次，而S组和NP组给予等容量生理盐水腹腔注射，连续给药至处死当天。S组、NP组和TMP组分别随机取6只大鼠，于术前1 d，术后3、7、14 d（T0~3）时测定机械痛阈（MWT）和热痛阈（TWL），并于T1~3时测定痛阈后处死，取术侧L4~L6背根神经节（DRG），采用实时荧光PCR法检测生长相关蛋白（GAP-43） mRNA的表达水平。
结果：与S组比较，NP组和TMP组MWT和TWL在T1~3时降低，背根神经节GAP-43 mRNA表达上调(P<0.05)。与NP组比较，TMP组MWT和TWL在T1~3时升高，背根神经节GAP-43 mRNA表达下调（P<0.05）。与T1时比较，NP组和TMP组MWT和TWL在T2~3时升高，背根神经节GAP-43mRNA表达下调（P<0.05）。
|Other Abstract||Objective：To evaluate the effect of tetramethylpyzine on the expression of GAP-43mRNA in dorsal ganglion root (DRG)in a rat model of chronic constriction injury, and to explore the curative effect and the involved mechanism of tetramethylpyzine in the neuropathic pain.Methods：Fifty-four healthy male Wistar rats weighing 180~220g and aging 6-8 w were randomly assigned to 3 groups(18 each): group sham operation(S); group neuropathic pain (NP) and group tetramethylpyzine(TMP). In group S, the right sciatic nerve was only exposed and in group NP and TMP, four loose ligatures were placed on the right sciatic nerve at 1mm interval with 4-0 silk suture. In group TMP tetramethypylrazine 100mgkg-1 everyday was administered after operation intraperitioneally until the day the rats was sacrificed while equal volume of normal saline was used to instead of tetramethylpyrazine in group S and NP. Mechanical withdrawl threshold (MWT) and thermal withdrawl latency (TWL) was measured at 1 day pre-operation(T0, baseline) and at 3, 7 and 14 days post-operation(T1~3). The animals were then killed. The L4~L6 dorsal root ganglions of chronic constriction injured sciatic nerves were removed for determination of the expression of GAP-43mRNA.Result：Compared with group S, the mechanical withdrawal threshold(MWT)and thermal withdrawal latency(TWL) was markedly reduced while the expression of GAP-mRNA was significantly increased at T1~3 in group NP and TMP(P<0.05).Compared with group NP, the MWT and TML was significantly increased and the GAP-43mRNA expression was markedly reduced in group TMP at T1~3(P<0.05). Compared with T1, the MWT and TWL was markedly reduced while the expression of GAP-43mRNA was significantly increased in group NP and TMP at T2~3(P<0.05).Conclusion：Intraperitioneal tetramethylpyrazine 100mgkg-1 everyday may inhibit hyperalgesia by reducing the expression of GAP-43 mRNA in dorsal root ganglion in a rat model of neuropathic pain.|
