| Abstract | 研究Twist2与E-cadherin在宫颈组织中的表达情况，探讨它们在宫颈癌变过程中的作用及意义。采用免疫组织化学法检测Twist2、E-cadherin在正常宫颈、CINⅠ、CINⅡ-Ⅲ、宫颈鳞癌4组中的表达情况及Twist2、E-cadherin在宫颈鳞癌中的表达与临床病理学特征的关系。应用Western blot法检测Twist2、E-cadherin在各级宫颈组织中的表达情况。结果显示：①免疫组织化学法显示：Twist2与E-cadherin在正常宫颈组织、CIN（Ⅰ)、CIN（Ⅱ-Ⅲ）、宫颈鳞癌中的阳性表达率分别为：6.7%、25.7%、47.7%、79.2%和93.3%、74.3%、52.3%、24.5%。各组之间比较,差异均有统计学意义(P<0.05)。Twist2、E-cadherin的阳性表达率在宫颈鳞癌的分化程度、临床分期、有无淋巴结转移的各组间差异有统计学意义（P＜0.05），在不同年龄间差异无统计学意义（P＞0.05）。②Western blot法显示: 对照组、CIN组、宫颈鳞癌组间Twist2逐渐升高，E-cadherin逐渐降低，三组间的相对表达量差异有统计学意义（P<0.05）。③Twist2与E-cadherin在宫颈鳞癌组织中的表达呈负相关（r=-0.401，P=0.003）。Twist2、E-cadherin可能共同参与了宫颈鳞癌的恶性转化过程, 联合检测Twist2与E-cadherin有助于判断宫颈癌的恶性程度。 |
| Other Abstract | Research Twist2 and E-cadherin expression in cervical tissue case and to explore their role in the process of cervical lesions and significance.Immunohistochemical assay Twist2, E-cadherin in normal cervical, CIN Ⅰ, CIN Ⅱ - Ⅲ, SCC expression in group 4 and Twist2, E-cadherin in cervical squamous cell carcinoma relationship with clinicopathological features . Application Western blot detect Twist2, E-cadherin at all levels of expression in cervical tissue. The results showed that: ① immunohistochemistry Show: Twist2 and E-cadherin in normal cervical tissue, CIN (Ⅰ), CIN (Ⅱ-Ⅲ), cervical squamous cell carcinoma, the positive expression rates were: 6.7%, 25.7%, 47.7 %, 79.2% and 93.3%, 74.3%, 52.3%, 24.5%. Among the groups, the differences were statistically significant (P <0.05). Twist2, E-cadherin positive expression rate in cervical squamous cell differentiation, clinical stage, lymph node metastasis of differences between groups was statistically significant (P <0.05), no difference between the different age groups was significant (P > .05). ② Western blot assay showed: the control group, CIN group, cervical cancer group Twist2 gradually increased, E-cadherin decreased, three relative expression levels between the two groups was statistically significant difference (P <0.05). ③ Twist2 and E-cadherin in cervical squamous cell carcinoma was negatively correlated (r = -0.401, P = 0.003). Twist2, E-cadherin may be involved in the process of malignant transformation of cervical squamous cell carcinoma, combined detection Twist2 and E-cadherin can help determine the degree of malignancy of cervical cancer. |
