Abstract | 目的 探讨PARP-1抑制剂AG014699在体外对人卵巢癌细胞株SKOV3增殖及凋亡所产生的影响,从而为卵巢癌的治疗寻找一种新的途径的可能,提高卵巢癌患者的治疗效果,提高她们的生存率。
方法 1.MTT:采用MTT法测定AG014699在1.0、10、20、50、100μmol/L等不同浓度分别作用24小时、48小时和72小时在体外对人卵巢癌细胞株SKOV3增殖作用的直接影响;从而根据MTT实验结果确定在下一步流式细胞仪检测中AG014699的作用浓度和作用时间。
2.流式细胞仪测定:用流式细胞仪测定AG014699在50μmol/L浓度下对人卵巢癌细胞株SKOV3作用24h后对细胞周期分布的改变及凋亡的影响。
结果 1.MTT测定:在24、48、72h时AG014699在10、20、50、100μmol/L浓度时分别测得的OD值与对照组相比P<0.05,有统计学差异;而随着AG014699作用时间的延长增加,不同浓度作用下的抑制率逐渐降低,各药物浓度作用在24h时抑制作用最强,IC50约为100μmol/L。
2.流式细胞仪测定:AG014699在50μmol/L浓度下对SKOV3作用24h能明显改变细胞的周期分布:与对照组的细胞相比,G0/G1期细胞所占比例明显增加(P<0.05);同时AG014699在50μmol/L浓度下能明显诱导卵巢癌细胞的凋亡。
结论 PARP-1抑制剂AG014699在体外能明显抑制卵巢癌细胞株SKOV3的增殖并能改变细胞的周期分布,诱导其凋亡。 |
Other Abstract | OBJECTIVE To explore the effects of Poly(ADP-ribose) Polymerase-1 Inhibitor AG014699 on the cell proliferation and apoptosis of human ovarian cell line skov3. Maybe can discover a new medicine to used in the patients of ovarian cancer, and improve the survival rate of ovarian cancer patients.
METHODS Growth inhibition rate of skov3 cells was measured with MTT method, at 24h、48h、72h of 1.0、10、20、50、100μmol/L, to determine the time and consistency usd in flow cytometery.Apoptosis rate and cell cycle distribution of the skov3 cells at 50μmol/L were inspected by flow cytometery after 24h.
RESULTS At 10, 20, 50,100μmol/L ,AG014699 inhibited the growth of human ovarian cancer cells skov3 at 24 48 and 72h, P<0.05, compared control group. The growth inhibition rate were increased with the growth of medicine’s consistency, especially at 24h.At 50μmol/L , AG014699 induced the apoptosis of the human ovarian cancer cell line skov3 at 24h, and changed the cell cycle distribution of skov3 cell compared control group ,increasd the rate of G0/G1, P<0.05.
CONCLUSION AG014699 significantly inhibited the growth of ovarian cancer cells skov3 and induces apoptosis. |
