Abstract第一部分 目的: 是探讨核因子-κB p50 (NF-κB p50) 和 P16INK4A 蛋白在HPV6/11阳性的尖锐湿疣(condyloma acuminatum, CA)组织中表达。方法: 采用荧光定量聚合酶链反应(Fluorescence quantitative Polymerase Chain Reaction, FQ-PCR)检测确定低危组CA即 HPV 6/11 阳性、HPV 16/18 阴性标本 21 例;免疫组织化学LDP 法检测 21 例低危组CA、 18 例正常包皮组织中 NF-κB p50 和P16INK4A的表达。结果: NF-κB p50阳性表达主要定位于细胞浆,部分细胞核也表达,NF-κB p50在正常包皮和低危组CA组织中阳性表达强度比较有统计学意义(Z=-4.412, P < 0.05);P16INK4A阳性表达主要定位于细胞浆,P16INK4A在正常包皮和低危组CA组织中阳性表达强度比较有统计学意义(Z=-2.669, P < 0.05);NF-κB p50 和P16INK4A在CA组织中表达无相关性(rs=0.009, P﹥0.05)。结论: NF-κBp50和P16INK4A表达可能参与了低危组CA组织细胞的增殖与调控。 第二部分 目的: 是探讨P16INK4A、NF-κB p50在宫颈尖锐湿疣(condyloma acuminatum, CA)、宫颈上皮内瘤变(cervical intraepithelial neoplasia CIN)、宫颈癌组织中的表达。方法: 采用免疫组织化学LDP 法对21例CA,24例CINI标本、10例CINll、15 例宫颈癌进行P16INK4A和NF-κB p50蛋白分析。结果: NF-κB p50在CA、CINI、CINll、宫颈癌中阳性表达率均为100%,但NF-κB p50在CA、CINI、CINll、宫颈癌组织中阳性表达强度比较有统计学意义(P < 0.05)。P16INK4A在CA阳性表达率为14.28%,在CINI、CINll、宫颈癌中阳性表达率均为100%,P16INK4A在CA、CINI、CINll、宫颈癌组织中阳性表达强度比较有统计学意义(P < 0.05)。结论: NF-κB p50可能参与了感染HPV16/18阳性组织的细胞增殖与调控;P16INK4A 蛋白主要在宫颈上皮内瘤变、宫颈癌组织中表达,故P16INK4A 蛋白有望作为一种肿瘤标记物用于癌前病变的筛查与诊断。
Other AbstractThe First Part Objective: To investigate the expression of NF-κB p50 and P16 INK4A in condyloma acuminatum(CA) with HPV6/11 positive. Methods: 21 cases of HPV 6/11 positive and HPV16/18 negative were diagnosed by Fluorescent quantitative PCR(FQ-PCR). The expression of NF-κB p50 and P16 INK4A were detected by immunohistochemistry technique with 21 cases of CA lesions and 18 cases of normal human skins(foreskin). Results: NF-κB p50 expression mainly localizes in the cytoplasm, few can be found to express in nucleus, The expression of NF-κBp50 in normal foreskin and 21 case of CA appears a significantly difference,(Z=-4.412, P< 0.05). P16 INK4A positive expression mainly localizes in the cytoplasm and the expression of P16 INK4A in the two groups also show a significantly difference (Z=-2.669, P < 0.05). There is no evidence to conform the relationship of the expression of NF-κB p50 and P16 INK4A in the lesions of CA. (rs=0.009, P0.05), Conclusions: NF-κB p50 and P16INK4A may be involved in the cell proliferation and control in the lesions of CA with HPV6/11 positive. The Second Part Objective: Expression of NF-κBp50 and P16INK4A in CA, CIN and cervical cancer.three tissues with HPV16/18 positive and HPV6/11 negative were diagnosed by in situ hybridization. Methods: Immunohistochemistry(LDP)to detect the expression of P16INK4A and NF-κBp50 in 21 cases CA, 24 cases CINI ,10 cases CINll and 15 cases cervical cancer patients respectively. Resuls: The positive rate of P16INK4A protein in CA was 14.28%,The positive rate in CIN and cervical cancer are both 100%. The expression of P16INK4A protein in CA, CINI, CINll and the cervical cancer had a significantly difference(p < 0.05). The positive rate of NF-κBp50 protein in CA, CINI, CINll and cervical cancer were 100% respectively, a The expression of NF-κBp50 protein in the group of CA, CINI, CINll and the cervical cancer show significantly difference (P < 0.05).Conclusions: Activated NF -κBp50 may play an role in human papillomavirus infection tissue .The positive expression of P16INK4A appears in CIN and cervical cancer, which means it can be take as a biomarker for CIN screening and diagnosis of ealier cancer.
