|Other Abstract||Object: To investigate the expression of MCM5 and P16INK4A in cervical tissues of differential pathological types, especially in cervical intraepithelial neoplasia(CIN) and cervical carcinomas, in order to analyse the relationship of MCM5 and P16INK4A with clinical and pathological features of cervical carcinoma and the relevance of each other.
Methods: Real-time fluorescence quantitative PCR and immunohistochemical staining SABC method were used respectively to examine the expression of mRNA and protein of MCM5 and P16INK4A in 40 cases of cervical squamous cell carcinoma, 11 cases of CINⅠ, 15 cases of CINⅡ-Ⅲ and 15 cases of normal cervical tissues. The expression of of mRNA and protein of MCM5 and P16INK4A was compared in different cervical tissues.
Results: 1. The expression of MCM5 and P16INK4A mRNA added with the progress of cervical lesion. The expression of mRNA of MCM5 was significantly up-regulated in carcinomas, CINⅠ and CINⅡ-Ⅲ than normal cervical tissues, there were significant difference with each other . There were significant difference when the group of cervical cancer compared with the group of CINⅠ and normal cervical tissues . MCM5 and P16INK4A positive expression rates in 15 normal cervical tissue,11 CINⅠ,15 CINⅡ-Ⅲ and 40 cervical squamous cell carcinoma samples were 20%, 63.6%, 73.3%, 97.5% , 0%、27.3%、86.7%、95.0% respectively. 2. there was association between MCM5 expression and clinical stage, the differentiation. however, there was no association between MCM5 expression and age. there was association between P16INK4A expression and the differentiation of cervical cancer, but the expression of P16INK4A showed no significant relation in different age and clinical stage. In invasive cervical cancer, the expression of MCM5 and P16INK4A showed positive correlation.
Conclusion: The expression of MCM5 and P16INK4A increased with the progress of cervical lesion. The expression of MCM5 and P16INK4A had correlation with the grades of histological in cervical squamous carcinoma. There was an important meaning of predicting outcome of cervical intraepithelial neoplasia and the progression and prognosis of cervical cancer by union examining MCM5 and P16INK4A .|
