Abstract目的 观察COX-2抑制剂对烫伤后大鼠COX-2及VEGF表达的影响。 方法 将90只SD大鼠随机分为A、B、C三组,其中A组为空白对照组(10只),B组为烫伤对照组(40只),C组为烫伤给药组(40只)。B、C组予以氯胺酮麻醉,10%的硫化钠脱毛,将大鼠背部浸入90℃水10s后造成面积为20%的深Ⅱ度烫伤模型。三组均给予正常饲料喂养,C组伤后给予COX-2抑制剂(塞来昔布)0.4mg/(g.d)共7d。B、C组于伤后1、3、5、7、14d取创面组织标本,A组在同样的时间点取背部组织标本。用免疫组化SABC法将观察标本中环氧化酶-2(COX-2)和血管内皮生长因子(VEGF)的表达情况。 结果 伤后1、3、5d,B组COX-2及VEGF的表达逐渐增高,C组COX-2及VEGF均有所下降(P<0.05);伤后7、14dB组COX-2表达下降而VEGF表达增加,C组与B组相比COX-2、VEGF的表达较低。 结论 COX-2抑制剂可抑制创面组织中COX-2、VEGF的表达,COX-2和VEGF在创面的愈合中发挥着重要作用。 关键词 烫伤;创面愈合;环氧化酶-2;血管内皮生长因子;环氧化酶抑制剂
Other AbstractAIM To observe the effect of COX-2 inhibitor on expression of COX-2 and VEGF in scalded rats. Methods 90 SD rats were randomly divided into group A, B and C. Group A was blank group(10) ;Group B was simple scald control group(40); Group C was COX-2-inhibitor treatment group(40). Group B and C were given to anesthetize by ketamine and depilate by 10% of the sodium sulfide. Then back of rats were immersed in 90 ℃ water for 10 seconds, and induce 20%TBSA of deep Ⅱ degree scaled model. Three groups were all given normal diet. Group C were given COX-2 inhibitor (Celecoxib west 0.4mg/(g.d), day1-7) . The back tissue samples of all Groups were harvested at day1,3,5,7,14 after scalded. To detect the cyclooxygenase -2 (COX-2) and vascular endothelial growth factor (VEGF) expression in all samples with immunohistochemical SABC method. Results The expression of COX-2 and VEGF in group B gradually increased during day1,3,5 after scald. Group C significantly decreased than that of group B (P <0.05); The expression of COX-2 in group B descended at day7,14 after scald, but the expression of VEGF still ascend at same time. Compared to group B, the expression of COX-2 and VEGF in group C was significant lower. (P<0.05) Conclusion COX-2 and VEGF play important roles in wound healing.COX-2 inhibitors could significantly inhibit the expression of COX-2, VEGF in scald tissue, Key words : scald; Wound healing ;cyclooxygenase -2 vascular endothelial growth factor; cyclooxygenase -2 inhibitor
