Abstract目的:探讨前列腺癌组织中CD146的表达与其微血管形成的作用及前列腺癌临床病理特征的关系。 方法 :采用免疫组织化学(EnVision+TM)方法检测38例前列腺癌和30例前列腺增生(BPH)组织标本中CD146、VEGF(血管内皮生长因子)的表达情况和CD34标记的MVD值(肿瘤微血管密度)。 结果:CD146在前列腺癌组织中阳性表达率为65.8%,在前列腺增生组织中CD146阳性表达率为13.3%,两组差异有显著性(P<0.01);前列腺癌不同分化程度、临床病理分期及骨转移的有无之间,CD146的表达差异有统计学意义(P<0. 05) 。前列腺癌组织中VEGF的阳性表达率为71.05%,前列腺癌组织MVD值为36.10±9.94,二者均较良性前列腺增生组数值明显升高(P<0.05);前列腺癌组织中CD146的表达与VEGF表达和MVD值均有相关性(P<0.05、r=0.518),前列腺癌组织中CD146表达越高,相应VEGF的表达越高,MVD值越大。CD146阳性组与CD146阴性组间的MVD值存在显著性差异﹙P<0.01﹚。 结论: CD146参与了前列腺癌浸润和转移过程;CD146可能在前列腺癌微血管形成过程中发挥了重要作用;CD146可作为判断前列腺癌预后的指标和治疗的新靶点。
Other Abstract"Background & Aims: CD146, also known as Mel-CAM, MUC18, is a membrane glycoprotein which functions as a Ca2+-independent cell adhesion molecule involved in heterophilic cell–cell interactions. CD146 is classified as a member of the immunoglobulin gene superfamily, involved in the process of angiogenesis and vasculogenesis of prostate cancer. CD146 has been demonstrated to be over-expressed in localized and metastatic prostate cancer, suggesting that it may be an important tumor marker. The purpose of this study was to investigate the Correlation between the expression of CD146 in prostate cancer tissue and angiogenesis of prostate cancer and it’s the clinicopathological characteristics significance. Methods: the expression of CD146 and VEGF in 38 specimens of prostate cancer tissue and 30 specimens of benign prostatic hyperplasia were detected with EnVisionTM immunohistochemical technique. Tumor microvessel density(MVD) was evaluated using anti-CD34 antibody as an endothelial marker with the same technique as well. Results: CD146 expression was positive in 4(13.3%) BPH specimens and 25 (65.8%) prostate cancers; CD146 protein expression was significantly increased in prostate cancers as compared to benign (P < 0.01). The expression of CD146 were correlated to differentiation degree, clinical staging, bone metastasis of prostate carcinoma (P<0.05). The positive expression rates of VEGF in the 38 cancer specimens were 71.05%, as compared with relatively lower rates in benign prostatic hyperplasia group(P<0.05),And the mean value of MVD was 39.10±9.94 , 25.40±10.25 respectively in prostate cancer , benign prostatic hyperplasia; The mean value of MVD was significant difference between benign prostatic hyperplasia and prostate cancer (P< 0. 01), The expression of CD146 were correlated significantly with the level of VEGF and tumor MVD in cancer tissues (P<0.05,r=0.518). Both the expression level of VEGF and the value of MVD increased when the expression level of CD146 in cancer tissues rised; Significant difference between in prostate cancers with CD146-positive expression and those with CD146-negative expression (P< 0. 01). Conclusions: CD146 may induce carcinoma into the development of metastasis and infiltration of prostatic carcinoma; CD146 plays a critical role in the process of angiogenesis and vasculogenesis of prostate cancer.
