AbstractCHR12 and CHR23, which are members of the SWI2/SNF2 chromatin remodeling ATP family, play an important role in the growth and development of Arabidopsis thalianaL. They are involved in a series of physiological processes that encompass embryonic development to flowering. We used the microscope, electron microscope and gene chip to understand the role of CHR12 and CHR23 in the apical meristem development of Arabidopsis. We used yeast two-hybrid sieve library to screen the proteins that might interact with CHR23 and verified by yeast two-hybrid and bimolecular fluorescence complementary experiments. Finally, we analyzed the molecular mechanism of enhanced drought resistance of chr12 chr23 by RNA-seq and qPCR. The double mutants of chr12 chr23 have obvious developmental defects and these defects can be restored. Genes that were differentially expressed in chr12 chr23 stem tip tissues participated in more than 20 biological pathways. At the same time, we found that CHR23 could bind to the drought-stress-related protein Di19. In addition, chr12 chr23 double mutants were highly tolerant of drought stress. In this study, we prove that CHR12 and CHR23 are not only necessary for the maintenance and differentiation of stem cells in the SAM and RAM, but also participate in the regulationof hypocotyl growth. In addition, they also play an important role in drought stress response.
